Transcriptome sequencing of human breast cancer reveals aberrant intronic transcription in amplicons and dysregulation of alternative splicing with major therapeutic implications

چکیده
After the publication of the article, the authors retrospectively noted that the name of Dr Dawn Jones, a postdoctoral researcher in the laboratory of Professor Christine Gosden, should have been added to the authors' list, whose contribution to the study greatly augmented the exploration of all the RNA products via adoption of the RNASeq (Illumina) methodology. All the existing authors agree to the inclusion of Dr Jones as a co‑author, with the exception of Derek Gardener, who has retired from the Institute and is no longer contactable by e-mail (although we are confident that he would not raise any objection). The correct author list (and affiliations) should therefore have been published as follows: Shiva Seyed Forootan1, Joe M. Butler1, Derek Gardener1, Dawn Jones1, Alison E. Baird1, Andrew Dodson1, Alistair Darby2, John Kenny2, Neil Hall2, Andrew R. Cossins2, Christopher S. Foster1,3 and Christine M. Gosden1 1Molecular Pathology Laboratory, Department of Molecular and Clinical Cancer Medicine, Institute of Translational Medicine, Faculty of Health and Life Sciences, University of Liverpool, Royal Liverpool University Hospital, Liverpool L69 3GA; 2Center for Genomic Research, University of Liverpool, Liverpool L69 7ZB; 3HCA International, Pathology Laboratories, London WC1E 6JA, UK. We regret the oversight of having failed to include Dr Dawn Jones as an author on this paper. Role of Aberrant Alternative Splicing in Cancer
Alternative splicing can alter genome sequence and as a consequence, many genes change to oncogenes. This event can also affect protein function and diversity. The growing number of study elucidate the pathological influence of impaired alternative splicing events on numerous disease including cancer. Here, we would like to highlight the significant role of alternative splicing in cancer biolog...
متن کاملRestoration of correct splicing in IVSI-110 mutation of β-globin gene with antisense oligonucleotides: implications and applications in functional assay development
Objective(s): The use of antisense oligonucleotides (AOs) to restore normal splicing by blocking the recognition of aberrant splice sites by the spliceosome represents an innovative means of potentially controlling certain inherited disorders affected by aberrant splicing. Selection of the appropriate target site is essential in the success of an AO therapy. In this study, in search for a splic...
متن کاملTranscriptome Profiling of the Cancer, Adjacent Non-Tumor and Distant Normal Tissues from a Colorectal Cancer Patient by Deep Sequencing
Colorectal cancer (CRC) is one of the most commonly diagnosed cancers in the world. A genome-wide screening of transcriptome dysregulation between cancer and normal tissue would provide insight into the molecular basis of CRC initiation and progression. Compared with microarray technology, which is commonly used to identify transcriptional changes, the recently developed RNA-seq technique has t...
متن کاملبررسی ترنسکریپتوم و تخمین بیان ایزوفرمهای سه ژن از مسیر پیامرسانی PI3K و FGFR در سرطان مثانه
Background: Aberrant pre-mRNA alternative splicing is a common event in cancer cells. Many abnormally spliced RNA variants have been observed in tumor cells and they can be used as biomarkers or therapeutic targets in new drug design. Increasing our knowledge in understanding the mechanisms of alternative pre-mRNA splicing for cancer-related genes and determination of cancer specific isoforms a...
متن کاملA Pan-Cancer Analysis of Alternative Splicing Events Reveals Novel Tumor-Associated Splice Variants of Matriptase
High-throughput transcriptome sequencing allows identification of cancer-related changes that occur at the stages of transcription, pre-messenger RNA (mRNA), and splicing. In the current study, we devised a pipeline to predict novel alternative splicing (AS) variants from high-throughput transcriptome sequencing data and applied it to large sets of tumor transcriptomes from The Cancer Genome At...
